A Biblioteca Setorial Dr. Paulo Bezerra da Escola Multicampi de Ciências Médicas do Rio Grande do Norte oferece uma variedade de produtos e serviços à comunidade acadêmica e externa. No entanto, enfrenta desafios em promover e dar visibilidade a esses recursos. Este estudo teve como objetivo desenvolver um aplicativo móvel para facilitar o acesso e ampliar a visibilidade dos produtos e serviços da biblioteca, promovendo uma interação mais eficiente com os usuários. A pesquisa teve caráter exploratório, aplicado e qualitativo, sendo conduzida como um estudo de caso. O aplicativo foi desenvolvido por meio do Glide Apps, incluindo recursos como busca de informações e canais de comunicação direta. A avaliação do aplicativo, realizada com 52 participantes, destacou sua praticidade, intuitividade e relevância como ferramenta de integração acadêmica. Conclui-se que o aplicativo atende às necessidades informacionais da comunidade, potencializando a visibilidade e a utilização dos serviços da biblioteca.Artigo STAT3/NF‐κB signalling disruption in M2 tumour‐associated macrophages is a major target of PLGA nanocarriers/PD‐L1 antibody immunomodulatory therapy in breast cancer(British journal of pharmacology, 2021-03-31) Cavalcante, Rômulo; Ishikawa, Uta; Silva, Emanuell; Silva Jr, Arnóbio; Araújo, Aurigena; Cruz, Luis; Chan, Alan; Araújo Jr, Raimundo; 0000-0001-6418-2454Background and Purpose Inflammation associated with the tumour microenvironment (TME) is critical for cancer development, and immunotherapeutic strategies modulating the immune response in cancer have been crucial. In this study, a methotrexate‐loaded (MTX) poly(lactic‐co‐glycolic acid)‐based (PLGA) drug nanocarrier covered with polyethyleneimine (Pei) and hyaluronic acid (HA) was developed and combined with an PD‐L1 antibody to investigate anti‐cancer and immunomodulatory effects in breast cancer TME. Experimental Approach Naked or HA‐coated PeiPLGA‐MTX nanoparticles (NPs) were assessed on 4T1 breast cancer cells grown in culture and in a mouse model of orthotopic tumour growth. Tumours were evaluated by qRT‐PCR and immunohistochemistry. The cell death profile and cell migration were analysed in vitro in 4T1 cells. Polarization of murine macrophages (RAW cells) was also carried out. Key Results Naked or HA‐coated PeiPLGA‐MTX NPs used alone or combined with PD‐L1 antibody modified the tumourigenic course by TME immunomodulation, leading to reduction of primary tumour size and metastases. STAT3 and NF‐κB were the major genes downregulated by NPs. In tumor‐associated macrophages (TAM) such regulation switched M2 phenotype (CD163) towards M1 (CD68) and reduced levels of IL‐10, TGF‐β and CCL22. Moreover, malignant cells showed overexpression of FADD, APAF‐1, caspase‐3 and E‐cadherin, and decreased expression of Bcl‐2, MDR‐1, survivin, vimentin, CXCR4 and PD‐L1 after treatment with NPs. Conclusion and Implications NPs‐mediated STAT3/NF‐κB signalling axis suppression disrupted crosstalk between immune and malignant cells, reducing immunosuppression and critical pro‐tumour events. It occurs in almost all patients who receive radiotherapy of the head and neck, as well as patients who undergo hematopoietic cell transplantation. The pathophysiology of OM is complex, and there is no effective therapy. The aim of this study was to evaluate the effect of dexamethasone-loaded poly(D,L-Lactic-co-glycolic) nanoparticles (PLGA-DEX NPs) on an OM model induced in hamsters. The NPs were synthesized using the emulsification-solvent evaporation method and were characterized by the size, zeta potential, encapsulation efficiency, atomic force microscopy, physicochemical stability, and the in vitro release. The OM was induced by the administration of 5-FU on the first and second days and mechanical trauma on the 4th day of the experiment. PLGA-DEX NPs were administered to treat OM. The animals were euthanized on the 10th day. Macroscopic and histopathological analyses were performed, measurement of malonaldehyde (MDA) and ELISA was used to determine the levels of IL-1β and TNF-α. Immunoexpressions of NF-κB, COX-2, and TGF-β were determined by immunohistochemistry, and qRT-PCR was used to quantify the gene expression of the GILZ, MKP1, and NF-κB p65. Walker 256 tumor is an excellent model for studying these changes associated with cancer in different organs, including injuries in testicular functions. However, the effects of supplementing glutamine on testicular morphometry in this model have not yet been investigated. Thus, the objective of this study was to evaluate the effect of L-glutamine supplementation on testicular morphometry in rats transplanted with Walker 256 tumor cells. Forty puberty Wistar rats were divided into four groups: control without L-glutamine (C); control supplemented with L-glutamine (CG); inoculated with Walker 256 tumor cells (WT) and inoculated with Walker 256 tumor cells and supplemented with L-glutamine (WTG). The testicles were removed, weighed, fixed in Bouin, and included in paraffin for histomorphometric analysis. Walker 256 tumor caused quantitative changes in the tubular and intertubular compartments and tunica albuginea, with reductions in the percentages of lumen and tunica albuginea, number of Sertoli cells per gram of testis; number of Leydig cells; percentage of blood vessels and connective tissue in intertubule. However, glutamine supplementation prevented part of these changes caused by the tumor, presenting mainly a protective effect on the tunica albuginea and percentage of blood and lymph vessels in the intertubule. Zirconia/hydroxyapatite (80/20) scaffold was characterized by X-ray diffraction (XRD) and Scanning Electron Microscopy (SEM). Critical size (8 mm) calvarial defects were created in wistar rats (n=48) and divided into four groups (90 days): G0 Group: positive control; G1 Group: hydroxyapatite; G2 Group: Zirconia; G3 Group: Zirconia/hydroxyapatite (80/20). Calvaria were subjected to Micro CT, histological and immunohistochemical analyses (RANK, RANKL, OPG, osteocalcin and FGF-2). IL-1 beta, IL-10 and TNF-alpha levels were analyzed by Elisa Immunoassay. The XRD analysis confirmed the formation of a crystalline structure and SEM showed the presence of regions corresponding to Zirconia and Hydroxyapatite. The Micro CT showed increased bone volume (BV/TV) and bone mineral density (BMD) in the G3 group (P<0.05). In addition, discrete periosteal bone formation was found at the interface of the defect edge and the external surface of the scaffold in the G3 group, showing osteocytes inside and osteoblasts (P<0.05) with scarce mononuclear inflammatory cells (P<0.01) in the central region of the defect. The immunostaining was moderate for RANKL, Osteocalcin and FGF-2 in the G3 group (P<0.5), while it was intense for OPG (P<0.001). IL-1 beta levels were decreased and IL-10 levels increased (P<0.05). One barrier to successful regeneration in the adult CNS is the diminished axonal growth capacity after maturation. Therefore, strategies that seek to promote the restoration of function to the chronically injured spinal cord have high therapeutic value. Neurotrophic factors and peripheral nerves are known to be good substrates for bridging the lesions associatedwith CNS trauma. The role of fibroblast growth factor-2, when added to the sciatic nerve, was examined following spinal cord injury in a rat. We evaluated whether FGF-2 added to a sciatic nerve graft placed in a gap promoted nerve recovery following a complete transection of the spinal cord and if it could enhance neuronal plasticity. Rats underwent a transection at the thoracic level, which was repaired with saline or a fragment of the sciatic nerve. In another group, FGF-2 was added immediately after thelesion. The effects of FGF-2 and the fragment of the sciatic nerve graft on neuronal plasticity were investigated at the epicenterof the injury using NF-200 and 5-HT immunoreactivity after 8 weeks.A high number of NF-200 and 5-HT immunoreactive fibers were observed in the treated groups with sciatic nerve graft in the presence or absence of FGF-2 when compared to the saline group. However, a small number of NF-200(p=0.03)and 5-HT fibers were observed in the epicenter of the graft when FGF-2 was added,when compared to the group that received sciatic nerve graft. This study aimed to evaluate morphometric and phenotypic changes in MCs grown with facial nerve-conditioned medium in the presence or absence of fibroblast growth factor 2 (FGF-2). For quantitative phenotypic analysis, the expression of GFAP, OX-42, MAP-2, β-tubulin III, NeuN, and NF-200 was analyzed by immunocytochemistry. Cells cultured with facial nerve-conditioned medium in the presence of FGF-2 expressed GFAP, OX-42, MAP-2, β-tubulin III, NeuN, and NF-200. On average, the area and perimeter of GFAP-positive cells were higher in the group cultured with facial nerve-conditioned medium compared to the group cultured with conditioned medium and FGF-2 (p=0.0001). Its synthesis is regulated by the enzyme called tyrosine hydroxylase. 3-hydroxytyramine/dopamine (DA) is a precursor of noradrenaline and adrenaline synthesis and acts as a neurotransmitter in the central nervous system. The three main nuclei, being the retrorubral field (A8 group), the substantia nigra pars compacta (A9 group) and the ventral tegmental area (A10 group), are arranged in the die-mesencephalic portion and are involved in three complex circuitries – the mesostriatal, mesolimbic and mesocortical pathways. These pathways are involved in behavioral manifestations, motricity, learning, reward and also in pathological conditions such as Parkinson’s disease and schizophrenia. The aim of this study was to perform a morphological analysis of the A8, A9 and A10 groups in the common marmoset (Callithrix jacchus – a neotropical primate), whose morphological and functional characteristics support its suitability for use in biomedical research. Coronal sections of the marmoset brain were submitted to Nissl staining and TH-immunohistochemistry. The morphology of the neurons made it possible to subdivide the A10 group into seven distinct regions: interfascicular nucleus, raphe rostral linear nucleus and raphe caudal linear nucleus in the middle line; paranigral and parainterfascicular nucleus in the middle zone; the rostral portion of the ventral tegmental area nucleus and parabrachial pigmented nucleus located in the dorsolateral portion of the mesencephalic tegmentum. The A9 group was divided into four regions: substantia nigra compacta dorsal and ventral tiers; substantia nigra compacta lateral and medial clusters. No subdivisions were made for the A8 group. These results reveal that A8, A9 and A10 are phylogenetically stable across species. The involvement of fibroblast growth factor-2 (FGF-2) activation in the dorsal root ganglion (DRG) was examined following spinal cord injury in the rat. We evaluated whether FGF-2 increases the ability of a sciatic nerve graft to enhance neuronal plasticity, in a gap promoted by complete transection of the spinal cord. The rats were subjected to a 4 mm-long gap at low thoracic level and were repaired with saline (Saline or control group, n = 10), or fragment of the sciatic nerve (Nerve group, n = 10), or fragment of the sciatic nerve to which FGF-2 (Nerve + FGF-2 group, n = 10) had been added immediately after lesion. The effects of the FGF-2 and fragment of the sciatic nerve grafts on neuronal plasticity were investigated using choline acetyl transferase (ChAT)-immunoreactivity of neurons in the dorsal root ganglion after 8 weeks. Preservation of the area and diameter of neuronal cell bodies in dorsal root ganglion (DRG) was seen in animals treated with the sciatic nerve, an effect enhanced by the addition of FGF-2. Thus, the addition of exogenous FGF-2 to a sciatic nerve fragment grafted in a gap of the rat spinal cord submitted to complete transection was able to improve neuroprotection in the DRG. The results emphasized that the manipulation of the microenvironment in the wound might amplify the regenerative capacity of peripheral neuronsArtigo Qualidade de vida e aspectos psicossociais da síndrome dos ovários policísticos: um estudo quali-quantitativo(Thieme Open, 2013-11) Moreira, Simone da Nóbrega Tomaz; Sá, Joceline Cássia Ferezini de; Costa, Eduardo Caldas; Azevedo, George Dantas deObjetivos: avaliar a qualidade de vida das mulheres com síndrome dos ovários policísticos (SOP) e compreender a experiência vivida por essas mulheres diante dos sintomas que apresentam. Métodos: este estudo compreendeu duas abordagens metodológicas - quantitativa e qualitativa, de forma complementar. Foi avaliada a qualidade de vida de 213 mulheres (abordagem quantitativa) por meio do SF-36, sendo 109 com SOP (Grupo Caso: 26,8±5,4 anos) e 104 mulheres saudáveis (Grupo Controle: 23,9±6,7 anos). A análise estatística compreendeu a utilização dos testes t de Student e qui-quadrado, além dos testes de correlação de Pearson. O nível de significância adotado foi de 5%. Das mulheres do Grupo SOP, 15 participaram do estudo qualitativo, tendo sido entrevistadas mediante uso de roteiro semiestruturado. Os dados qualitativos foram analisados por meio da técnica análise de conteúdo temática categorial. Resultados: mulheres com SOP apresentaram comprometimento na qualidade de vida quando comparadas ao Grupo Controle (capacidade funcional: 76,5±20,5 e 84,6±15,9, respectivamente; aspectos físicos: 56,4±43,3 e 72,6±33,3; estado geral de saúde: 55,2±21,0 e 62,5±17,2; vitalidade: 49,6±21,3 e 55,3±21,3; aspectos sociais: 55,3±32,4 e 66,2±26,7; aspectos emocionais: 34,2±39,7 e 52,9±38,2; saúde mental: 50,6±22,8 e 59,2±20,2). Em relação aos dados qualitativos, a análise temática categorial aponta que sentimentos de "anormalidade", tristeza, medo e ansiedade estiveram associados aos principais sintomas da SOP: hirsutismo, irregularidade menstrual, infertilidade e obesidade. Esses sintomas repercutiram na vida social, na esfera profissional e no relacionamento conjugal dessas mulheres. Conclusão: a SOP compromete a qualidade de vida das mulheres, levando-as a se sentirem diferentes das outras mulheres. Methods: tumor necrosis factor-alpha (TNF-a), interleukin-6 (IL-6) and C-reactive protein (CRP) were assessed in serum samples from 40 women in reproductive age. Patients were divided into four groups: I (non-eutrophic with PCOS, n¼12), II (non-eutrophic without PCOS, n¼10), III (eutrophic with PCOS, n¼8) and IV (eutrophic without PCOS, n¼10) being non-eutrophic, the overweight and obese women with BMI425 kg/m2 and eutrophic women with BMI524.9 kg/m2. Homeostatic model assessment (HOMA-IR) was calculated for evaluation of IR. Results: PCOS patients showed increased levels of CRP (p50.01) and HOMA-IR (p50.01). When divided by BMI, both non-eutrophic and eutrophic PCOS showed increased CRP levels (p50.01) and HOMA-IR (p50.01). There were no differences in TNF-a and IL-6 levels between groups. Methods: semi-structured, questionnaire-based interviews were conducted with 285 women who underwent spontaneous abortion and 31 who reported having induced it. The data were analyzed using the thematic analysis technique, and, subsequently, by the IBM SPSS Statistics Standard Edition software program. The significance level was set at p < 0.05. Results: in both capitals, the women who induced an abortion referred to the partner as the person who could not find out about the abortion (p<0.01 in Natal; p = 0.02 in São Paulo-SP) and, simultaneously, as the one who could have avoided it (p < 0.01 in Natal; p = 0.03 in São Paulo). In Natal-RN, induced abortion was associated with the partner’s absence at the time pregnancy was confirmed (p = 0.02) and, in Sao Paulo-SP, with their negative reaction to news of the pregnancy (p = 0.04) and lack of participation in the abortion process (p < 0.01). Conclusion: despite having achieved independence, women still regard male participation in the abortion process as an important factor. Métodos: foi desenvolvido instrumento de avaliação cognitiva em saúde sexual e reprodutiva com base nos temas abordados no componente curricular optativo Saúde Reprodutiva, resultando em prova objetiva de múltipla escolha contendo 27 itens. Os temas selecionados foram: direitos humanos, sexuais e reprodutivos (DHSR), sexualidade, violência institucional, gênero, violência sexual, concepção, contracepção, aborto/interrupção legal da gestação, mortalidade materna e doenças sexualmente transmissíveis (DSTs) – HIV/AIDS. Os temas foram agrupados em três dimensões do conhecimento: DHSR, legal/institucional e biomédica. Na análise de covariância, dois modelos multivariados foram ajustados. Resultados: participaram do estudo 183 alunos, 127 do grupo que cursou o componente curricular eletivo saúde reprodutiva (Grupo SR) e 56 do grupo que não cursou (Grupo Não SR). Noventa e seis alunos (52,5%) eram do sexo masculino e 87 (47,5%) do sexo feminino. A média de idade foi de 24,7±1,9 anos no Grupo SR e de 24,4±2,6 no Não SR. O desempenho médio do Grupo SR foi superior ao Não SR nos temas DHSR, sexualidade, violência institucional, violência sexual, aborto/interrupção legal e DSTs – HIV/AIDS. Não houve diferença no desempenho dos sexos masculino e feminino, com a exceção do tema mortalidade materna, no qual o grupo masculino foi inferior (6,9±0,2 e 7,8±0,2, respectivamente; p<0,05). Métodos: estudo exploratório, quantitativo, realizado na Maternidade Escola Januário Cicco, em Natal. A amostra foi de 130 mulheres, 65 com pré-eclâmpsia e 65 normotensas. Resultados: constatou-se diferença estatística significativa no que se refere ao índice de massa corpórea, peso, histórico familiar de doenças cardiovasculares e complicação cardiovascular entre as mulheres com pré-eclâmpsia prévia e as normotensas. Os grupos desconheciam os fatores de risco cardiovasculares e, além disso, referiam dificuldades de acesso aos serviços de atenção primária. Many studies have been conducted about efficacy of UL treatment, but little research has been performed about womens health related quality of life (HRQL). Methods: this is a semi-structured, descriptive, observational, qualitative study that was performed during eight months. Focus group (FG) interviews were performed with women attending at a tertiary hospital in Brazil, who were consecutively included in the study. Seventy women with symptomatic UL were recruited to this study. FG duration was one hour with mediators with 5-6 women at each group. Collected data from discussions was processed according to thematic analysis and stored at a qualitative software. Results: women were negatively influenced by the presence of symptomatic UL. The major themes that were noticed during analysis were: beliefs and attitudes towards UL; limitation to social and professional activities; sensation of fear/unfairness/discouragement towards the symptoms and adverse effects during treatment with GnRH analogs. Conclusions: symptomatic UL has a negative impact on womens HRQL. Health providers should consider such impact when counseling women on their treatment options, since it may have an important influence in these patients’ decision-making process. The aim of this study was to analyze if overweight/obese women with polycystic ovary syndrome (PCOS) meet the American College of Sports Medicine (ACSM) recommendation regarding to exercise intensity to improve health status during a single bout of ‘‘ARE". Methods: a sample of 14 overweight/obese women with PCOS (18–34 years) performed a single bout of ‘‘ARE’’ (40 min of aerobic exercise on outdoor track). The Feeling Scale (FS) was used to guide ‘‘ARE’’ intensity/pace maintaining an affective valence between ‘‘good’’ and ‘‘very good’’ during all time. Heart rate (HR), speed, % of HR at first and second ventilatory threshold (VT1 and VT2) and time spent at moderate (64–76% of HRmax) and vigorous (77–95% of HRmax) intensity during ‘‘ARE’’ were measured with a global positioning system (GPS) device. Results: volunteers exercised at 73% (68–78%) of HRmax, 5.8 (5.2–6.2) km/h, 93.4% of HR at VT1 (89.3–98.2) and 80.5% of HR at VT2 (75.3–84.6) and spent 480% of time at moderate intensity. Conclusions: overweight/obese women with PCOS met the ACSM recommendation regarding exercise intensity to improve health status when exercised between ‘‘good’’ and ‘‘very good’’ of FS. Métodos: participaram do estudo mulheres selecionadas aleatoriamente, na fase do climatério, residentes na cidade de Natal, Rio Grande do Norte, localizada na região do nordeste brasileiro. Foram utilizados os questionários UQOL e SF-36, sendo a fase da tradução realizada da língua inglesa para o português por três professores, enquanto que a fase de adaptação da versão traduzida foi feita através da aplicação do questionário a 35 mulheres, que poderiam marcar a opção de resposta “não compreendi a questão”; e para a validação foram usadas as medidas de reprodutibilidade (teste-reteste) e validade de construto, seguindo as normas metodológicas padronizadas internacionalmente. Resultados: a versão brasileira foi reconhecida plenamente pela população-alvo, que foi composta por 151 mulheres, devido a nenhuma questão apresentar percentual de “não compreensão” igual ou superior a 20%. Os resultados obtidos para a reprodutibilidade intra e interobservador demonstraram concordância significativa em todos os itens do questionário. Essa versão apresentou consistência acima do critério requerido (>70), demonstrando sua precisão, enquanto que a validade de construto foi obtida através de correlações estatisticamente significativas entre os domínios ocupação, saúde e emocional do UQOL com os domínios do SF-36. O coeficiente alfa de Cronbach para o instrumento como um todo foi de 0,82, representando boa precisão. Análise da correlação item-total demonstrou a homogeneidade da escala. Conclusão: a partir das etapas realizadas, o questionário UQOL foi traduzido e adaptado para aplicação no Brasil, apresentando alta reprodutibilidade e validade. Dessa forma, pode ser incluído e utilizado em estudos brasileiros que visem avaliar a qualidade de vida durante a peri e pós-menopausa
